纳米壳寡糖-铁配合物的制备及其生物活性的研究

Study on the Preparation and the Biological Activity of Nano Chitosan-oligosaccharide/Fe Complex

用分子量为5000的壳寡糖与FeCl3反应制得纳米壳寡糖-铁配合物, 并通过FTIR, UV, DTA, TEM, PCS等手段对其结构与形态进行了表征. 同时用紫外光谱法和凝胶电泳法对纳米壳寡糖以及纳米壳寡糖-铁配合物的生物活性进行了研究, 实验结果表明纳米壳寡糖-铁配合物是以静电嵌插的方式与质粒DNA进行结合, 而致使DNA损伤.

The nano chitosan-oligosaccharide/Fe complex was prepared by addition of FeCl3 to chitosan-oligosaccharide solution, the molecular weight of which was about 5000. The complex was characterized by FTIR, UV spectra and DTA, TEM and PCS method. It was concluded that the Fe(III) was coordinated with the NH2 of chitosan-oligosaccharide. And the ultraviolet spectroscopy analysis and gel electrophoresis analysis were used to research the biological activity of chitosan-oligosaccharide and chitosan-oligosaccharide/Fe complex via plasmid DNA. The result indicated that the chitosan-oligosaccharide/Fe complex had a strong binding affinity with DNA and the complex could damage and degrade DNA.
