Effects of TiO2 Nanoparticles on the Photodamage of CT-DNA by Ru(II) Bis(terpyridine) Complexes

Due to their short excited state lifetimes, Ru(II) bis(terpyridine) complexes can hardly used as DNA photocleavers. In this paper three Ru(II) bis(terpyridine) complexes [Ru(ttp)(tpy)]2+ (1), [Ru(ttp- COOH)(tpy)]2+ (2) and [Ru(ttp-COOH)(tpy-pyr)]2+ (3) were designed and synthesized (tpy=2,2':6',2"- terpyridine, ttp=4'-(p-tolyl)-2,2':6',2"-terpyridine, ttp-COOH=4'-(p-carboxyl-phenyl)-2,2':6',2"-terpyridine, tpy-pry=4'-(1-pyrenyl)-2,2':6',2"-terpyridine). It was found that TiO2 nanoparticles improved the photodamage ability of 3 on calf thymus DNA in both aerated and deaerated conditions. The oxidized form of 3, [Ru(III)(ttp-COOH)(tpy-pyr)]3+, generated by photoinduced electron transfer between TiO2 nanoparticle and 3, was invoked to account for the enhanced DNA photodamage.
