Study on the Synthesis, DNA Interaction, and Solvatochromism of a Hydroxyethyl Carboxylate-Grafted Dipyridophenazine Ruthenium(II) Complex

A new ruthenium(II) polypyridyl complex of [Ru(bpy)2(hedppc)](ClO4)2 {in which bpy=2,2'-biyridine, hedppc=2-hydroxyethyl dipyrido[3,2-a:2',3'-c]phenazine-11-carboxylate} was synthesized and characterized by elemental analysis, 1H NMR and MALDI-TOF MS. The binding of the com-plex to calf thymus (ct) DNA was studied with UV-visible absorption and luminescence spectrophotometric titrations, DNA competitive binding with ethidium bromide, viscosity measurements and thermal denatura-tion experiments. The results indicated that the complex was bound to ct-DNA through an intercala-tive mode with an intrinsic binding constant of (6.99±1.34)×106 L/mol in buffered 50 mmol/L NaCl comparable to parent [Ru(bpy)2(dppz)]2+. The complex is per-manently non-emissive in the absence and presence of the DNA, and exhibits intriguing solvatochromic properties different from parent [Ru(bpy)2(dppz)]2+.
