The aim of the study is to determine the role of nicotine as a lipase inhibitor in fat and fatty acid metabolism and to explain the clinic phenomenon: smokers weigh less than age-matched nonsmokers, and most of smokers gain weight after smoking cessation. Lipase activity was measured in the presence or absence of nicotine using olein as substrate. Lipase is active in vitro. Nicotine is demonstrated to be an uncompetitive inhibitor of lipase at biologically reachable concentration. It is shown that the activity of lipase was reduced to 50% at lipase concentration of 7 mg/mL.

Key words: lipase, nicotine, olein, oleic acid
