Micro-plate Magnetic Chemiluminescence Immunoassay of Carbohydrate Antigen 125 in Serum

A micro-plate magnetic chemiluminescence immunoassay (MMCLIA) for the determination of human carbohydrate antigen 125 (CA125) was developed based on traditional micro-plate chemiluminescence immunoassay (micro-plate CLIA) and magnetic particle-based chemiluminescence immunoassay (MP-CLIA). This method used magnetic particles as solid support and the micro-plate as a reactor. The horseradish peroxidase (HRP)-luminol-H2O2 chemiluminescent system was chosen as the detection system. The results showed that the developed method had a detection limit of 2.0 U/mL with a linear range of 0~400 U/mL. The developed method showed wider linear range than the micro-plate CLIA. The sensitivity and precision were higher than the commonly used MP-CLIA method. In addition, the method has been successfully applied to detection of CA125 in human serum with good correlation compared with the Roche method.
