A novel and efficient synthesis of thiochromene-containing spiro pyrrolidine oxindoles has been developed via a 1,3-dipolar cycloaddition reaction. In this transformation, (E)-2-styryl-4H-thiochromen-4-one serves as the dipolarophile, while azomethine ylides generated in situ from isatins and L-proline act as the 1,3-dipoles. Notably, the cycloaddition selectively occurs at the γ,δ-double bond remote from the electron-withdrawing carbonyl group, preserving the conjugated thiochromenone framework-a scenario rarely reported in such conjugated systems. The three-component one-pot reaction proceeds under mild conditions to afford a broad range of spirooxindoles in moderate to good yields with high regio- and chemoselectivity. The synthetic utility is demonstrated by a gram-scale experiment and subsequent derivatizations, including N-acylation and selective oxidation of the sulfur atom to the corresponding sulfoxide. This work expands the chemical space of spirooxindole-based scaffolds and provides a practical strategy for constructing sulfur-containing spiro heterocycles with potential bioactivity.
