本文报道用高锰酸钾在碱性溶液中氧化3-甲硫基-5-羟基-6-甲基-1,2,4-三嗪(3)以合成3-甲砜基-5-羟基-6-甲基-1,2,4-三嗪(13),后者在缓和条件下经亲核性取代反应制得一系列3-取代-5-羟基-6-甲基-1,2,4-三嗪类化合物(1)。为研究1,2,4-三嗪类化合物生理活性,拟合成(1)类化合物。3-巯基-5-羟基-6-甲基-1,2,4-三嗪(2),3-甲硫基-5-羟基6-甲基-1,2,4-三嗪(3),3-乙硫基-5-羟基-6-甲基-1,2,4-三嗪(4)及3-苄硫基-5-羟基-6-甲基-1,2,4-三嗪(5)均已合成[1,2]

The syntheses of some 3-substituted-5-hydrozy-6-methyl-1,2,4-triazines (1) are reported.The oxidation of 3-methylthio-5-hydrogy-6-methyl-1,2,4-triazine (3) with potassium permanganate at room temperature in aqueous sodium hydroxide solution yielded 3-methylsulfon-5-hydroxy-6-methyl-1,2,4-triazine (13) which reacted with nudeophilio reagents (e.g.ammonia,hydrazine hydrate,P-toluidine,isobutylamine,piperidine,sodium ethoxi.de,sodium phenoxide) under mild conditions to give the corresponding 3-amino-5-hydroxy-6-methyl-1,2,4-triazine (6),3-hydrazine-5-hydroxy-6-methyl-1,2,4-triazine (7),3-P-toluidino-5-hydroxy-6-methyl-1,2,4-triazine (8),3-iso-butylamino-5-hydroxy-6-methyl-1,2,4-triazine(9),3-piperidino-5-hydroxyl-methyl-1,2,4-triazine (10),3-ethoxy-5-hydroxy-6-methyl-1,2,4-triazine (11) and 3-phenoxy-5-hydroxy-6-methyl-1,2,4-triazine (12).
