Enantioseparation of Chiral Compounds on Amylose-tris(cyclohexylcarbamate) Chiral Stationary Phase by High-performance Liquid Chromatography

Amylose-tris(cyclohexylcarbamate) (ACHC) coated on a kind of small particle silica gel was prepared. On this chiral stationary phase (CSP), the direct enantiomeric separation of nine chiral compounds has been achieved using n-hexane as the mobile phase containing various alcohols as modifiers. The effect of mobile phase modifiers on their resolution and retention was investigated. Chromatographic behavior of chiral compounds on ACHC-CSP was compared with that on amylose-tris(3,5-dimethylphenylcarbamate) by hexane/isopropanol as the mobile phase. The result suggests that ACHC showed high chiral recognition abilities as well as amylose-tris(3,5-dimethylphenylcarbamate) in HPLC. ACHC-CSP is a suitable CSP for the optical resolution of a lot of racemates by HPLC.
