Studies on the Oxidation of σ-Phenylenediamine Catalyzed by Chloroperoxidase in the Presence of an Ionic Liquid as Co-solvent

The oxidation of σ-phenylenediamine (OPD) catalyzed by chloroperoxidase (CPO) in the presence of an ionic liquids (ILS) as co-solvent was studied in this work. Identifications by UV-Vis, FT-IR and 13C and 1H NMR spectrum show that the product is 2,3-diaminophenazine (DAP). This ensures the application of (OPD-H2O2-CPO) system in fluorescence enzyme-link immunoassay. The enzyme kinetic assay indicated that both the affinity and specificity of CPO to the substrate was improved in the buffer-ILS mixture, which was the main reason of the enhancement of product yield. This enzyme-catalyzed reaction was controlled mainly by the stability and consumption of CPO. The DAP yield was 81.16% under the optimum condition.
