Effect of pH, Quercetin and Zinc Ion on the Structure of β-Casein by Circular Dichroism Method

The effect of pH, quercetin (Qct) and Zn(II) on the structure of β-casein (β-CN) in solution were investigated using circular dichroism spectroscopy. The results indicated that the secondary structures of β-CN in the solution were induced by the pH, quercetin and Zn(II)-Qct complexes. In pH 7.6 PBS buffer, β-CN contain 32.6% α-helix, 53.5% β-sheet and 13.9% γ-turn and random coil, the addition of Qct cause increase the content of α-helix and decrease the content of β-sheet, while Zn(II) and Zn(II)-Qct complexes lead to formation of γ-turn and random coil with the decease of α-helix.
