Progress in the Total Synthesis of Tumor-Associated Carbohydrate Antigen Globo-H Hexasaccharide

Tumor-associated hexasaccharide Globo-H is one of the complex carbohydrate antigens anchored on the surface of many cancer cells,such as the prostate and breast cancer cell。Its discovery has led opportunities for the development of carbohydrate-based vaccine and cancer immunotherapy,though the chemical preparation with high purity and efficiency still challenges the synthetic community。The review summarizes the total synthesis progress of Globo-H since its first synthesis by Danishefsky’s glycal assembly strategy in 1995。Other state-of-the-art technologies include Schmidt’s trichloroacetimidate methodology,Boons’ two-directional glycosylation strategy,Wong’s reactivity based one-pot method,Seeberger’s linear solution-phase synthesis and automated solid-phase synthesis,Huang’s multi-component one-pot synthesis as well as the latest enzymatic synthesis。In conclusion concerning the synthetic methodology,thioglycoside is a versatile precursor,glycal,trichloroacetimidate and fluoroglycoside are widely applied,and phosphate glycosyl donor is prominent in solid-phase synthesis。These strategies represent the significant advances and trends of current carbohydrate chemistry.
