本文报道了带保护基的四核苷三磷酸(Bz)AbzpbzUbzpUbzpCBzbz,(9),(Bz)AbzpbzUbzpCbzpbzU(Bz2(15),CbzpbzGibupacGibupacA(Bz)2(bz)2(24)和四核苷酸(Bz)-AbzpbzUbzpUbzp Cvbzpbz(29)的合成。三个四核苷三磷酸均用相应的二核苷二磷酸和二核苷一磷酸,即化合物9由(Bz)AbzpbzUbzp(4)和UbzpC(Bz)2bz(8),化合物15由4和CbzpbzU(Bz)2(14),化合物24由(McOTr)CbzpbzGibupac(19)和GibupacABz2(bz)2(23)缩合而得。四核苷酸(Bz)AbzpbzUbzpUbzpCbzpbz(29)由化合物4和二核苷酸Ubzpbz(28)缩合而成。四者均用二环己基碳二亚胺为缩合剂。化合物9、15、24和29在去保护基、纯化后分别得到纯的ApUpUpC(10)、ApUpCpU(16)、CpGpGpA(25)和ApUpUpCp(30)。在反应后处理条件和二乙基氯乙基纤维素柱层析条件下,对化合物Gpac、Gpibu、Gacpac、Gibupac、Gbzpac和Gibupibu上的酰基保护基的稳定性作了比较。

The tetraribonucleoside triphosphates ApUpUpC(10),CpGpGpA(25) and the tetranucleotide ApUpUpCp(30) corresponding to 58-61 and 62-65 fragments of the yeast alanine transfer ribonucleic acid molecule together with another tetraribonucleoside triphosphate ApUpCpU (16) have been synthesized.As shown in Fig.1,2,3,and 4,protected tetranucleoside triphosphates and tetranucleotide were obtained by condensation of the appropriately protected dinucleotide with protected dinucleoside monophosphates or protected dinucleotides.The protected dinucleotides and dinuc;leoside monophosphates were prepared by coupling the appropriate protected nucleotide with proteoted nucleo ride and nucleoside respectively.
