微流控芯片电泳快速分离脂蛋白

Rapid Separation of Lipoproteins Based on Microfluidic Chip Electrophoresis

描述了一种芯片电泳快速分离脂蛋白的方法. 利用自制的微流控芯片及激光诱导荧光技术电泳分离经硝基苯并噁二唑-C6-酰基鞘胺醇预染的脂蛋白标本, 在40 mmol/L tricine缓冲液(pH 9.4)中加入40 mmol/L甲基葡胺, 在500 V电压下40 s进样, 在2000 V 电压下2 min内完成分离, 可出现低密度脂蛋白(LDL)与高密度脂蛋白(HDL)两条脂蛋白区带, 5次重复性试验其出峰时间变异系数(CV)为2.6%. 本法为高血脂患者提供了一种快速、简便、灵敏、重复性好的诊断方法.

A rapid assay of separating lipoproteins with microfluidic chip is described. The pre-stained lipoproteins with nitrobenzoxadiazole-6-ceramide were separated and analysed in quartz chip with laser induced fluorescence (LIF) detector. The electrophoresis buffer was accomplished with 40 mmol/L tricine buffer by addition of 40 mmol/L methylglucamine. After 40 s sample injection by applying 500 V voltage, the 2000 V separation voltage was switched on, and the peaks of low-density lipoprotein (LDL) and high-density lipoprotein (HDL) were detected within 2 min. After calculation coefficient of variation (CV) of sample migrating time was not more than 2.6% through 5 times detection. The method was sensitive, rapid, simple and reproducible for diagnosis of hyperlipoidemia.
