毛细管电泳电致化学发光法测定家犬体内甲氧氯普胺的血药浓度

Determination of Metoclopramide in Dog Plasma by Capillary Electrophoresis with Electrochemiluminescence

基于甲氧氯普胺对钌联吡啶电化学发光信号的增敏作用, 建立了检测甲氧氯普胺的毛细管电泳电致化学发光新方法. 最佳实验条件为: 检测电位1.18 V, 钌联吡啶浓度5 mmol/L, 检测池磷酸缓冲溶液40 mmol/L (pH 7.2), 分离磷酸缓冲溶液20 mmol/L (pH 6.1), 进样电压12 kV, 进样时间10 s, 分离电压14 kV. 方法的检测限(3σ)为5.0×10-3 mg/L, 线性范围为 0.03~9.52 mg/L, 相关系数为0.9990, 回收率为98.0%~101.7%. 对家犬体内甲氧氯普胺血药浓度的监测发现, 给药1.5 h血药浓度达到最大值, 血药浓度的半衰期约为4 h. 本方法具有简便、快速、灵敏、进样量少等特点, 可用于甲氧氯普胺血药浓度监控和药物动力学研究.

Based on the phenomenon that metoclopramide (MCL) could consumingly enhance the electrochemiluminescence (ECL) intensity of tris(2,2 -bipyridyl)ruthenium , a novel and sensitive method was proposed for determination of metoclopramide using capillary electrophoresis (CE) coupled with electrochemiluminescence (ECL) detection. The influence of several experiment parameters was explored. The optimum experimental conditions were detection potential 1.18 V (vs. Ag/AgCl), 40 mmol/L of phosphate buffer (pH 7.2) containing 5 mmol/L in CEL detection cell, running buffer of 20 mmol/L (pH 6.1), sample injection of 10 s at 12 kV, and separation voltage of 14 kV. The detection limit (3σ) of this method was 5.0×10-3 mg/L. The linear range of ECL intensity versus concentration of metoclopramide was 0.03~9.52 mg/L, and the correlation coefficient was 0.9990. The recovery of testing 5 standard samples by this method was 98.0%~101.7%. Maximum of MCL concentration in Dog Plasma appeared at 1.5 h of drugging, and half life of the concentration was about 4 h. This method had the merits of simpleness, speediness, sensitivity, little sample injection, and could successfully avoid the influencing of interference. This method was successfully utilized to the assay of metoclopramide in dog plasma.
