氨基糖的研究 Ⅳ.N-(2’,4’-二硝基苯基)-氨基糖的高碘酸氧化

STUDIES ON AMINO SUGAR Ⅳ.PERIODIC OXIDATION OF N-DNP-AMINO SUGARS

我们曾进行N,N'-双二硝苯基链胺(Ⅰ),N-二硝苯基-2-胺基-2-脱羟-β-D-葡萄吡喃糖甲甙(Ⅱ)及N-二硝苯基-2-胺基-2-脱羟-α-D-葡萄吡喃糖甲甙(Ⅲ)的高碘酸氧化.在二昼夜内(Ⅰ)消耗1.9分子的高碘酸,(Ⅱ)及(Ⅲ)分别消耗0.99及1.2分子.自(Ⅰ)的氧化反应中获得其氧化产物,经证明该化合物是一种由二醛化合物和一分子水缩合而成吡喃环状的半缩二醛化合物.上述结果证明与羟基相邻的胺基经二硝苯基保护后,不再参与高碘酸氧化反应,而这种基团的存在对其他1,2-二醇的高碘酸氧化反应并无影响.

Periodic oxidation of N,N‘-bis-DNP-streptamine (I) (DNP=2,4 dinitrophenyl), methyl N-DNP-β-D-glucosaminide (II) and methyl N-DNP-a-D-glucosaminide (III) has beerm studied.In aqueous methanol, the compounds I, II and III consumed 1.9, 0.99 and 1.2 moles of periodic acid respectively within 48 hrs.A product isolated from the periodic oxilotion of I was recrystallized from methanol- or ethanol-petroleum ether and found by analysis to contain one molecule of the corresponding alcohol in its composition.It showed no aldehydic carbonyl adsorption in both IR and UV spectra.Acetylation of the oxidatiozn product yielded a triacetate derivative (VI).Thus the recrystallized oxidation products mentioned above must exist in the cyclic "hemialdal"[10,11] form, shown as (Va) and (V6) respectively, in spite of the fact that a bis-p-nitrophenylhydrazone has been obtained froera Va.It can also be concluded that the N-DNP group neither participates in nor gives any.significant effect on the oxidative splitting of the a-glycol by periodic acid,
