醇酮环合法合成dl-麝香酮

麝香酮(β-甲基环十五烷酮)是天然麝香的组成成分,并具有天然麝香某些重要的药理作用,本文报道它的合成工艺和纯化的研究。有关麝香酮的合成方法,国内外报道很多[1]。我们选用α,ω-长链二元酸酯进行醇酮环合法制备dl-麝香酮。早先Stoll和Commarmont[2]报道用β-甲基十五烷二酸双乙酯经醇酮环合法合成dl-麝香酮。

dl-Muscone was prepared through acyloin cyclization of dimethyl β-methyl-pentadecanedioate,which was synthesized from anodio coupling of half methyl ester of dodecanedioic acid and that of,β-methylglutaric aoid.Separtion of dl-muscone from its isomerio γ-methyl compound was achieved by silica-gel column chromatography,The result shows that ratio between dl-muscone and its γ-methyl isomer is about 7:3.
