DNA-binding Spectrum and Photo-cleavage of Polypyridyl Ruthenium(II) Complex on DNA

Agarose gel electrophoresis was used to study the binding and photo-cleavage of pBR322 plasmid DNA with polypyridyl ruthenium(II) complex [Ru(bpy)2(ODCIP)]2+ (bpy: 2,2 -dipyridyl, ODCIP: 3,4- dichloro-imidazo[4,5-f][1,10]phenanthroline). Since the polypyridyl ligand ODCIP has multi coordination sites, the coordination of [Ru(bpy)2(ODCIP)]2+ with Zn2+ and their binding and photo-cleavage effect on DNA were also investigated by UV-vis absorption spectroscopy, fluorescent spectroscopy and agarose gel electrophoresis experiments. The results showed that the complex had a good photo-cleavage effect and might produce a combination of superoxide anion radicals and singlet oxygen mechanism for the DNA scission. The binuclear complex [Ru(bpy)2(ODCIP)Zn]4+ could be formed by [Ru(bpy)2(ODCIP)]2+ and Zn2+, and it bound to DNA by intercalation but the photo-cleavage of pBR322 plasmid DNA had not been notably enhanced.
