光子晶体薄膜的制备以及在编码载体中的应用

Fabrication of Photonic Crystal Films and Their Application in Encoded Carriers

通过将由聚苯乙烯纳米粒子构成的光子晶体膜镶嵌在聚二甲基硅氧烷(PDMS)薄膜中制备得到了具有PDMS/光子晶体/PDMS夹心结构的可用于多重生物分析的光子晶体编码载体. 用编码载体进行了大肠杆菌3种基因的杂交检测: 以3种光子晶体膜作为编码载体固定核酸探针, 然后在含有荧光标记的目标分子的缓冲液中进行杂交反应. 杂交反应后以光子晶体膜的特征反射谱为核酸编码, 以荧光信号的有无来确定目标分子的存在与否. 实验结果表明PDMS/光子晶体/PDMS夹心结构是一种有效的构建悬浮载体的方法.

Encoded carriers with a polydimethylsiloxane (PDMS)/photonic crystal/PDMS sandwich structure were fabricated, which could be used for multiplex bioassay. Photonic crystal film was derived by self-assembling monodisperse polystyrene nanoparticles. Detection of three kinds of genes of E. coli was demonstrated with the use of the carriers. These encoded carriers were firstly immobilized with different nucleic acid probes. Then the nucleic acid probes on the carriers were hybridized in the buffer containing target molecules tagged with fluorescent molecules. After hybridization the target molecules were detected by fluorescence and the codes of the carriers were identified by the reflection spectra. Experimental results showed that PDMS/photonic crystal/PDMS sandwich structure was a good approach for the construction of the suspended carriers.
