磁性纳米氧化铁与小牛胸腺DNA相互作用研究

Study on the Interaction between Magnetic Iron Oxide Nanoparticles with Calf Thymus DNA

以微波辅助共沉淀法合成磁性纳米氧化铁(MNPs), 分别采用紫外-可见吸收光谱(UV-vis)、拉曼光谱(Raman)及琼脂糖凝胶电泳(GE)研究其与小牛胸腺DNA (ct DNA)的相互作用. UV-vis显示出明显的增色效应, 这是由DNA分子受MNPs的干扰碱基外露而引起的|Raman研究表明DNA分子碱基、脱氧核糖以及磷酸骨架均受到MNPs一定程度的干扰|而GE证实了MNPs并未使DNA分子发生断裂、双链解开等本质性变化. 该研究对MNPs在生物医学领域的安全应用具有重要意义.

Magnetic iron oxide nanoparticles (MNPs) were synthesized by microwave-assisted coprecipitation. The effects of MNPs on the calf thymus DNA (ct DNA) were investigated by UV-vis spectra, Raman spectra and Agarose gel electrophoresis, respectively. UV-vis spectra revealed the hyperchromic effect on ct DNA due to the exposure of basic group that induced by MNPs. Raman spectra indicated that the basic group, ribodesose and backbone were disturbed in the presence of MNPs. However, Agarose gel electrophoresis confirmed that DNA melting or DNA strand break have not occurred under the influence of MNPs. The research had great significance for the safety application of MNPs in bio-medical field.
