对1株红树来源真菌Penicillium janthinellum XLN32122的次级代谢产物进行研究, 从中分离得到1个新的含有1,2-氧杂十二环的杂环二肽化合物trichodermamide H (1), 3个类似物以及3个已知的聚酮类化合物. 综合运用一维、二维核磁共振波谱、高分辨质谱和电子圆二色性(ECD)光谱确定了化合物1的平面结构和绝对构型. 体外抗炎活性结果显示, Trichodermamide B (3)对脂多糖(LPS)诱导的RAW 264.7细胞中NO的产生具有显著的抑制活性, IC50值为(13.13±0.005) μmol/L, 强于阳性对照药物地塞米松[IC50=(136.84±1.33) μmol/L]. 体外抗菌活性测试表明, 化合物3对耐甲氧西林金黄色葡萄球菌有一定的抑制活性(IC50=12.5 μg/mL), 阳性对照万古霉素的IC50值为1.56 μg/mL.

A new heterocyclic dipeptide with a highly functionalized 1,2-oxazadecaline core, named trichodermamide H (1), and three known analogues, along with three known polyketides, were isolated from the fermentation extract of the mangrove- derived fungus Penicillium janthinellum XLN32122. The structure of 1 was elucidated on the basis of extensive 1D and 2D NMR spectra data analysis, HR-ESI-MS, electronic circular dichroism (ECD) calculations. Trichodermamide B (3) exhibited better inhibitory effect on nitric oxide (NO) production in lipopolysaccharide (LPS) induced RAW 264.7 cells with an IC50 value of (13.13±0.005) μmol/L than that of the positive control dexamethasone [IC50=(136.84±1.33) μmol/L]. Compound 3 exhibited antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA) with an IC50 value of 12.5 μg/mL, while the positive control vancomycin showed an IC50 value of 1.563 μg/mL.
