稀土、钨酸、1,3-二苯基-4-吡唑甲醛缩异烟肼三元配合物的合成、表征及抗癌活性的研究

Synthesis, Characterization and Anticancer Activity of Rare Earth Complexes with 1,3-Diphenyl-4-pyrazole-formyl Isoniazidylhydrazone and Tungstic Acid

通过回流冷凝法和低温固相法制备了5种稀土、钨酸、1,3-二苯基-4-吡唑甲醛缩异烟肼三元配合物, 并通过元素分析, 摩尔电导, IR, UV, TG-DTA, TC-ICP, XPS等对其结构和化学组成进行表征, 确定其化学组成 Na[RE(HL)(WO4)2]•6H2O (HL: 1,3-二苯基-4-吡唑甲醛缩异烟肼). 采用MTT比色法研究了配合物对K562肿瘤细胞的细胞毒性, 结果表明: 三元配合物的IC50在25 μg/mL左右, 并进行了形态学观察. 流式细胞术结果表明: 配合物对癌细胞最佳凋亡浓度为12.5 μg/mL, 细胞被配合物阻滞在S期, 抑制了细胞的有丝分裂过程.

Using a reflux condensation method and a low-temperature solid phase method, five kinds of new ternary complexes were synthesized with rare earth chloride, tungstic acid and 1,3-diphenyl-4-pyrazole- formyl isoniazidylhydrazone. Their compositions were characterized by elemental analysis, molar conductance, FT-IR, UV-vis, TG-DTA, TC-ICP, and XPS, and confirmed to be: Na[RE(HL)(WO4)2]•6H2O (HL: 1,3-diphenyl-4-pyrazole-formyl isoniazidylhydrazone). MTT was used to study the complexes cytotoxicity against to K562 tumor cells and morphological observation. The results show that the complexes IC50 were 25 μg/mL, flow cytometry results showed that the best concentration apoptosising on cancer cells were 12.5 μg/mL. Cancer cells were arrested in S phase, inhibitting cells mitosis process.
