可逆识别Hg2+荧光探针的合成及活细胞中成像的应用

Synthesis of a Reversible Fluorescence Probe for Hg2+ and Its Application in Living Cells

以罗丹明B与邻苯二甲酰氯为原料缩合反应得到新化合物T1,通过荧光光谱法研究了化合物与金属离子的识别特性. 结果表明荧光探针T1能对Hg2+进行快速、选择性、可逆的识别,且常见金属离子对其干扰较小. 探针的荧光强度与Hg2+浓度(4×10-8~9×10-8 mol/L)呈现较好的线性关系. 探针在HepG2活细胞中的荧光显微成像表明T1可应用于生物体的检测.

A new rhodamine B derivative T1 has been rationally designed and synthesized facilely by the condensation reaction of rhodamine B hydrazide and o-phthaloyl chloride. The recognition properties of the target compound T1 with metal ions had been investigated by the fluorescence spectrophotometry. This probe exhibits several favorable features, including instantaneous and reversible recognition, high sensitivity and selectivity to mercury ions. When the concentration of Hg2+ was in the range of 4×10-8~9×10-8 mol/L, there was a good linearity between the fluorescence intensity and the concentration of Hg2+. The fluorescence imaging experiments of Hg2+ in HepG2 living cells demonstrated their valuable application in biological system.
