羟喜树碱(HCPT)是一种广谱的抗肿瘤药物, 对其具有识别作用的荧光分子不但可以用于检测, 还可因药物递送性能的提高而获得更高的活性. 因此, 寻找高选择性识别HCPT的荧光主体分子具有研究价值, 但具有一定挑战性. 合成了两亲性杯咔唑大环(3), 利用荧光光谱、核磁共振、扫描电镜和透射电镜等方法研究了其与HCPT的主客体间相互作用. 结果表明, 化合物3可经荧光共振能量转移效应实现比率型荧光选择性识别HCPT, 化学计量比为2∶1, 结合常数为1.26×1014 (mol/L)-2. 检测限(Limit of detection, LOD)为11.7 nmol•L-1. 此外, 化合物3的存在显著增强了HCPT体外抑制人乳腺癌细胞(MCF-7)细胞增殖活性, IC50值降低了两个数量级, 表明化合物3兼具分子荧光探针和药物分子载体的双重功能, 为后续深入研究提供科学依据.

The fluorescent molecule capable of recognizing 10-hydroxycamptothecin (HCPT), a broad-spectrum anti-tumor drug, is not only able to detect it, but also able to enhance its activity due to the improvement of the delivery. Therefore, the construction of highly selective fluorescent hosts of HCPT is valuable, but remains challenging. In this work, an amphiphilic carbazole macrocyclic molecule (3) was synthesized and the host-guest interaction between 3 and HCPT was studied by fluorescence spectroscopy, NMR, scanning electron microscopy and transmission electron microscopy. The results showed that 3 could recognize HCPT with a stoichiometric ratio of 2∶1 via the mechanism of fluorescence resonance energy transfer (FRET) and a binding constant of 1.26×1014 (mol/L)-2. The detection of limit (LOD) is 11.7 nmol•L-1. In addition, the complexation enhanced the activity of HCPT to inhibit the proliferation of MCF-7 (Michigan Cancer Foundation-7) cells in vitro, and the IC50 value decreased by two orders of magnitude, indicating that 3 possessed the dual function of both fluorescence probe and molecular carrier, thus providing scientific basis for further research.
