通过对药用植物仙鹤草(Agrimonia pilosa)内生链霉菌YINM00048的液体发酵产物的系统探究, 共分离获得10个化合物, 包括2个结构新颖的戊二酰亚胺类衍生物2-羟基-3-[2-(2-羟基-3-甲基-5-羟甲基苯基)-2-氧代乙基]戊二酰亚胺(1)和2-羟基-3-[2-(2-羟基-3-羟甲基-5-甲基苯基)-2-氧代乙基]戊二酰亚胺(2). 通过核磁共振(1D/2D NMR)、高分辨质谱(HR-ESI-MS)、单晶X射线衍射等技术的综合应用, 结合含时密度泛函理论-电子圆二色谱(TDDFT-ECD)计算法, 确证了化合物12的平面结构和绝对构型. 体外活性评价显示, 化合物2对人肺癌细胞A549和人肝癌细胞SMMC-7721具有较弱的抑制活性, IC50值分别为62.69和55.07 μmol/L.

Two new glutarimide derivatives, 2-hydroxy-3-[2-(2-hydroxy-3-methylphenyl-5-hydroxymethyl)-2-oxoethyl] glu- tarimide (1) and 2-hydroxy-3-[2-[2-hydroxy-3-hydroxymethyl-5-methylphenyl]-2-oxoethyl] glutarimide (2), along with 8 known compounds were isolated from the liquid fermentation broth of endophytic Streptomyces sp. YINM00048 obtained from Agrimonia pilosa. Their structures were elucidated through comprehensive spectroscopic analysis, including nuclear magnetic resonance, high-resolution mass spectrometry, and single-crystal X-ray diffraction, supported by time-dependent density functional theory (TDDFT)-electronic circular dichroism (ECD) calculations. Furthermore, compound 2 displayed weak cytotoxic activity against A549 and SMMC-7721 cell lines with IC50 values of 62.69 and 55.07 μmol/L, respectively.
