A Mimetic Peptide with Two Pairs of Concentrated Disulfide Bridges

A small linear peptide of 11 amino acids (NTLCCEGCMCY-COOH) has been designed and synthesized based on cysteine framework (CC—C—C) of α-conotoxins and some amino acid residues of natural Conus imperialis conotoxin. In CH3COONH4 or NH4HCO3 buffer, this peptide form only one oxidative product with disulfide connectivity “C(1)—C(4), C(2)—C(3)” which is different from main forms of α-conotoxins [C(1)—C(3), C(2)—C(4)]. This mimetic α-conotoxin peptide possesses analgesic activity and con-tains most concentrated disulfide bridges reported to date, and could be used as drug design template.
