31P NMR spectroscopy was applied to study the reaction course of the hydrolysis of UpU by RNase A. For the 1st time, the individual changes in UpU, U>p (uridine 2',3'-cyclophosphate), and Up (uridine 3'-phosphate) were continuously monitored and quant. determine The 2-step mechanism is thus incontestably proved. The rate of transphosphorylation (k1) is calculated to be ~40.9 s-1, and the rate for the hydrolysis of U>p (k2) is 9.4 s-1.
