用计算机辅助再设计了烟草、小麦和水稻的核糖酮-1,5-双磷酸羧化酶/氧化酶的成熟亚基(rbcS)的三个基因,结合化学方法和酶促配位进行了全合成,并在大肠杆菌中以高得产表达.

Three structural genes which code for the mature small subunit (rbcS) of ribulose-1,5-bisphosphate carboxylase/oxygenase (Rubisco) from tobacco, wheat, and rice, resp., were redesigned with the aid of a computer, totally synthesized by combining chem. methods and enzymic ligation, and expressed in E. coli with high yields. The genes were designed to express the codon usage preferred by the host cell and to include as many unique restriction sites as possible, including EcoRI and XbaI sites at the 5' and 3'-termini of the gene to enable easy transfer from one vector to another.
