抗体酶催化Oxy-Cope重排反应的研究

Polyclonal antibody with catalytic activity for oxy-cope rearrangement

A polyclonal antibody was generated against an antigen, a conjugate of carrier protein(BSA) and 2-[(p-(3-carboxypropyloxy)-phenyl]- cyclohexanol-1(3) (a transition state analogue of Oxy-Cope rearrangement), synthesized from cyclohexene in 7 steps. The antibody was found to catalyze the rearrangement of (3S, 4R)-3-p- methoxyphenyl-4-hydroxy-1,5-hexadiene (1) to trans-6-p-methoxyphenyl-5- hexe nal-1 (2) with ν~m~a~x=15 μmol/L·min, K~M=1025 μmol/L and K~c~a~t~.=10 min^-^1 in the presence of 0.2 mol/L MES[2-(N-Morpholino) ethanesulfonic acid] and 0.2 mol/L NaCl(pH=6.0 buffer) at 37℃ with the mole ratio of (3S, 4R)-3-p-methoxyphenyl-4-hydroxy-1,5-hexadiene (1) to polyclonal antibody being 100 : 1.
